Retatrutide
LY3437943 · triple agonist
Supplied in 10 mg and 30 mg vials.
Retatrutide is a synthetic triple agonist at the GIP, GLP-1 and glucagon receptors. What the published work at each of those three receptors examines, and where the trial programme stands, is set out under How it works below.

How it works
Of the three receptors this compound acts at, one is where Ozempic and Wegovy (semaglutide) act and two are where Mounjaro (tirzepatide) does. Glucagon is the third, and the one that has not appeared in an approved drug of this class — which is why the compound is studied on its own terms rather than as another incretin analogue. Material on this page is not those medicines.
GLP-1
Incretin receptor
The receptor behind the approved incretin drugs. The literature at it concerns appetite signalling in the central nervous system and glucose-dependent insulin release. That is what the papers examine, not a finding about this vial.
GIP
Second incretin receptor
The receptor a dual agonist adds. Studied for its part in how ingested energy is handled, and for how agonism there interacts with GLP-1 agonism in the same molecule. That handling is the research question; it is not a finding about this vial.
Glucagon
The third arm
Studied in relation to hepatic glucose output and to energy expenditure. This is the receptor that distinguishes a triple agonist from a dual one, and the least characterised of the three in this combination. Those are questions the glucagon literature asks, not findings about this vial.
Where the evidence stands
Phase 2 results in obesity and in type 2 diabetes were published in the New England Journal of Medicine in 2023, and the phase 3 TRIUMPH programme is still running. It remains investigational: no regulator has completed a review.
Laboratory results
